Prof. Varda Rotter (varda.rotter@weizmann.ac.il) and Prof. Moshe Oren (moshe.oren@weizmann.ac.il) are responsible for the establishment of immortalized primary smooth muscle and epithelial prostate cultures. Well characterized primary prostate cultivated cells will be obtained from the laboratory of H. Klocker and will be infected by hTERT viral stocks. All expression vectors to be used, will be constructed and prepared as viral stocks by Weizmann group. As a second phase of the study, expression of wild type p53 will be inactivated in the immortalized prostate cell cultures. Comparison of gene expression patterns of the various established cultures will be done by the use of Affymetrix DNA microarray Chips. Specific gene signature that are altered following p53 inactivation will be identified and characterized.

Track record:
Prof. Varda Rotter and Prof. Moshe Oren are studying p53 for more than 20 years. They have established active laboratories in the p53 field and gained experience in the molecular and cellular biology of p53. The laboratories have all the facilities and experience required for molecular biology, tissue culture experimentation, including cell immortalization. The Weizmann Institute has a central Affymetrix microarray facility, which has its own specialists both in the use of the microarrays and in the bioinformatics and data analysis.
